Production of light-coloured, low heat-absorbing Holstein Friesian cattle by precise embryo-mediated genome editing

چکیده

Context Genome editing enables the introduction of beneficial sequence variants into genomes animals with high genetic merit in a single generation. This can be achieved by introducing primary cells followed producing live animal from these somatic cell nuclear transfer cloning. The latter step is associated low efficiencies and developmental problems due to incorrect reprogramming donor cells, causing welfare concerns. Direct fertilised one-cell embryos could circumvent this issue might better integrate improvement strategies implemented industry. Methods In vitro zygotes were injected TALEN editors repair template introduce known coat colour dilution mutation PMEL gene. Embryo biopsies screened polymerase chain reaction sequencing for intended biallelic edits before transferring verified recipients development term. Calves genotyped their coats scanned visible hyperspectral cameras assess thermal energy absorption.Key results Multiple non-mosaic calves precision edited genotypes produced, including parents. Compared controls, showed strong which was lower absorbance. Conclusions Although biopsy screening not absolutely accurate, non-mosaic, precisely readily produced embryo-mediated editing. lighter colouring caused radiative heat gain help reduce stress.Implications study validates putative causative rapidly adapt grazing cattle changing environmental conditions.

Reproductive Performance for Holstein Friesian × Arsi and Holstein Friesian × Boran Crossbred Cattle

The study was carried out at Agarfa ATVET College dairy farm to evaluate the reproductive performance of Holstein Friesian× Arsi and Holstein Friesian ×Boran cattle. For the study, records compiled from 1983 to 2012 at the Agarfa dairy farm were used as original data. The effects of breed, bloodlines, season and parity on the reproductive traits were evaluated. Data were analyzed using the gene...

Genetic Association Analysis of Paratuberculosis Forms in Holstein-Friesian Cattle

A genetic susceptibility to Mycobacterium avium subsp. paratuberculosis (MAP) infections in ruminants has been longtime suspected to exist. Recently, natural infections in cattle have been reclassified into latent and patent forms based on histopathological findings and their associations with immunological and microbiological variables. This study aims to explore whether these newly defined ph...

A Genome Wide Association Scan of Bovine Tuberculosis Susceptibility in Holstein-Friesian Dairy Cattle

BACKGROUND Bovine tuberculosis is a significant veterinary and financial problem in many parts of the world. Although many factors influence infection and progression of the disease, there is a host genetic component and dissection of this may enlighten on the wider biology of host response to tuberculosis. However, a binary phenotype of presence/absence of infection presents a noisy signal for...

Differential macrophage function in Brown Swiss and Holstein Friesian cattle

There is strong evidence that high yielding dairy cows are extremely susceptible to infectious diseases, and that this has severe economic consequences for the dairy industry and welfare implications. Here we present preliminary functional evidence showing that the innate immune response differs between cow breeds. The ability of macrophages (MØ) to kill pathogens depends in part on oxygen-depe...

Mycobacterium bovis Infection in Holstein Friesian Cattle, Iran

To identify strains of Mycobacterium bovis circulating in Iran, we used region of difference, spoligotypes, and variable number tandem repeats to genotype 132 M. bovis isolates from Holstein Friesian cattle. Despite wide geographic origins, the strains were genetically homogeneous. Increased distribution of cattle herds and inadequate control measures may have contributed to strain dispersion.
